The only way to be sure that the hole is true, not just centered in each end, is to put it there by either method of assembly or cutting.

And if the drill bit wandered, which I would expect, a 1/2" threaded rod would not pass through and no way then to straighten out the hole.  I would expect a drill bit to follow an existing slot or channel without significant wandering if the bit was close to the size of the slot or channel.

That's the idea.
The drill will not wander if the pilot point isn't in contact with any wood. it would follow the predetermined path of the existing hole. You wouldn't technically be drilling but rather reaming out and existing hole by removing any glue squeeze out.
